Spheroid-on-Chip: Unraveling the Impact of Fluid Dynamics on Mesenchymal Stem Cell Spheroids
Presented by Dr. Apurva Mishra, Faculty of Dentistry, National University of Singapore
ibiTea Topic Outline
In the 11th ibiTea, Apurva Mishra will discuss the impact of microfluidics on the growth and differentiation of stem cells in a spheroid-on-chip model.
Specifically, the talk will focus on the effects of fluid flow on tooth-derived mesenchymal stem cells (MSCs) within a spheroid-on-chip microperfusion array. The investigation highlights the influence of fluid dynamics on the viability of MSCs within the spheroids and their ability to form bone-like tissue. The potential to control the spatio-temporal differentiation of MSCs within the spheroids by manipulating flow conditions is demonstrated, providing new insights for periodontal and bone regenerative therapies.
Representative confocal z-sections of spheroids after 14 days of culture under osteodifferentiation medium and stained with calcium-binding agent BoneTag (red) and DAPI (blue). The panels represent superficial and deeper z-sections of the spheroids to visualize the calcium deposits within peripheral and deeper zones (scale bar: 100 µm).
