HEK293 cells expressing GPCR NK-I on the outer cell membrane and cameleon YC3.6 in the cytoplasm. Fluorescently labelled NK-I ligand (SP-TAMRA) is added to the cells, resulting in a bright red glow of the cell membrane. Upon receptor activation calcium is released from intracellular stores inducing a change in the fluorescence properties of the calcium sensor cameleon: CFP excitation yields simultaneous CFP and YFP emissions by a FRET phenomenon. In overlap they here appear as a green glow in the cytoplasm.
