Transmigrating leukocyte through the endothelial barrier: A primary neutrophil is caught in the act. The neutrophil is migrating through two adjacent endothelial cells (HUVECs). HUVECs were electroporated with ICAM-1-GFP and RFP-LifeAct constructs two days prior to the experiment. HUVECs were treated overnight with TNF-alpha. Sample was fixed using 3.7% formaldehyde and permeabilized using 0.5% Triton X-100. In green, ICAM-1-GFP is visualized and in red, RFP-LifeAct can be seen. Neutrophil is visualized in blue. DIC image showed neutrophils as well as endothelial cell monolayer. Image was taken with a Zeiss 510 laser scanning confocal microscope.
