All-in-One Open Well Formats for IF

Cells are cultured in the classic open well geometry. Fixation, per­meabilization, and staining steps are also done in these wells. After staining and mounting, the sample can be observed through the coverslip-like bottom using high resolution microscopy.

The ibidi μ-Slide 8 well and the μ-Dish 35 mm, high allow the use of standard immunofluorescence protocols, without using coverslips, in an all-in-one open well chamber.

The ibidi Mounting Medium, which is specially designed to work with ibidi µ-Slides and µ-Dishes, prevents photobleaching and allows the sample to be stored for an extended period of time without the need for additional coverslips.

Limitations of All-in-One Open Well Formats

  • Storage time restricted to several weeks due to gas exchange through the plastic material from which the slides and dishes are made
  • Higher amounts of staining solution needed


Open Slides versus Channel Slides

In open formats, meniscus formation is prevented by using small coverslips. In channel structures this type of “lid” is integrated into the slide leading to perfect imaging quality without meniscus formation.
